An efficient micropropagation protocol was developed for Solanum caripense Dunal, a
native wild species with an encouraging biotechnological interest due to their edible fruits,
which contain appreciable quantities of vitamin C and sucrose that represent important
compounds in nutritious industry. In Stage 1, seeds were surface sterilized applying four
disinfection treatments (TD), and sowed for 42 days on M&S medium supplemented with
sucrose and activated charcoal; the best results were obtained from TD1 (Alcohol 30% +
NaClO 30% + H2O2 7%), which did not register any contamination and reached 92% of
germination. During Stage 2, stem segments of in vitro germinated seeds were subcultured
and exposed to nine multiplication treatments (TM) for 35 days, using M&S medium
supplemented with sucrose, activated charcoal and different concentrations of AIA and
BAP as phytoregulators; the best results were induced by TM4 (AIA 0.5 mg/l + BAP 0.5
mg/l) that produced 8.50 shoots/explant, and TM1 (AIA 0.0 mg/l + BAP 0.5 mg/l) with
13.88 nodes/explant. In Stage 3, the formed plantlets from multiplication phase were
subcultured for 42 days on peat containing IBA in different concentrations as ex vitro
rooting treatments (TE); finally, there were optimal results for rooted plants because all
treatments, TE0, TE1, TE2, TE3 and TE4 (0.0, 0.5, 1.0, 2.0 and 3.0 mg/l IBA,
respectively), achieved 100% of rooted plants |
