In recent years conservation research conducted by different naturalists and scientists have focused on preserving the flora, since it is a fundamental part of balanced ecosystems. In Ecuador, the level of endemism of Orchidaceae is high, so investigate alternative conservation is an important tool in order to prolong and ensure their permanence. This study was based on the development of a cryopreservation protocol seed four orchid Elleanthus Odontoglossum, Epidendrum and Oncidium; by evaluating the interaction of cryoprotectants following: C1 (1,5ml DMSO 1M), C2 (0,75ml DMSO 1M + 0,75ml Sucrose 1M), C3 (0,75ml DMSO 1M + 0,75ml Glycerol 1M), C4 (0,5 DMSO 1M + 0,5ml Sucrose 1M+ 0,5ml Glycerol 1M) and two cooling techniques: Fast (submerged directly to -196 °C); and shoulder (temperature drop to -80 °C and subsequently in liquid nitrogen at -196 °C). The plant material was obtained from the genebank of Orchids of Ecuador Phase I (section Cotacachi-Cayapas) stored in the research and biodiversity assessment CIVABI-UPS. The results show that in the rapid cooling technique, is the best cryoprotectant C1 (Dimethylsulfoxide), with the largest percentage viability tetrazolium test (ex vitro) in Elleanthus gender, Oncidium and Epidendrum averaging 66, 3%, 57,9% and 47,5% respectively. While in Odontoglossum C2 (Dimethylsulfoxide + Sucrose) with stepwise cooling technique, showed the best results with a 66,55% viability. For variable rate turgor (in vitro) the optimal seed storage protocol for all genders was more rapid technique C1 (Dimethylsulfoxide), presenting a range of turgor from 55% to 83,3%. |
