The genus Gaiadendron belonging to the Lorantaceae family, in Ecuador the Gaiadendron punctatum (Ruiz y Pav.) G. Don, is considered a native plant. This species was ancestrally used as a remedy for cough. The object to present this research was to establish an in vitro cultivation using seeds and sprouts of Gaiadendron punctatum (Ruiz y Pav.) G. Don, by the development of three phases. Analyze the concentration and immersion time of reagents for disinfection to prevent phenolization, were examined, after disinfection protocols were probed, and finally the MS salts concentration and temperature were valued. In the first phase, adequate concentrations of fungicide, acetic acid, citric acid, ethyl alcohol and sodium hypochlorite were determined for the prevention of bud phenolization. For the standardization of a disinfection protocol for buds, the applied treatments were 100% effective against fungal and bacterial contamination, whereas for seeds the TPS2 treatment proved to be the optimal treatment for fungal contamination. In the stage of introduction and adaptation to the cultivation medium it was determined that at a concentration of 50% of mineral salts of MS cultivation medium and at a temperature of 20 ° C of incubation, 100% germination of seeds is obtained and favors the Obtaining of seedlings of greater length, therefore the use of seeds for the in vitro propagation of this species is recommended. |
