The following research work was conducted with the purpose of isolating and identifying yeasts of the Saccharomyces gender from the tomato tree fruit (Solanum betaceum), by macroscopic, microscopic and biochemical identification analysis in addition to screening tests, through the use of the selective culture medium Lysine.
Two strains of Saccharomyces cerevisiae were identified from the 57 samples of mature tomato tree fruit, corresponding to 6% of total isolated yeast strains, and the remaining 94%, corresponding mainly to Rhodotorula, Candida and Cryptococcus genders.
After isolation and identification, the Saccharomyces cerevisiae strains were exposed in YPD medium supplemented with 6.0, 8.0 and 10.0° GL ethanol, demonstrating 100% growth of the strain exposed to GL 6.0°, 90% growth at 8.0 ° GL, and 60% growth at a concentration of 10.0° GL after 96 hours of incubation. These results were obtained after comparing the growth masses with that of the main strain, which was isolated in a culture medium without the presence of ethanol and incubated under similar temperature conditions.
The selected strains of Saccharomyces cerevisiae, Rhodotorula mucilaginosa, Candida albicans, Cryptococcus neoformans and Cryptococus neoformans var. uniguttulatus were placed in conservation vials CRIOBANK® and frozen at -80°C. |
